Abstract

Plant tissure culture is a technique to grow the plant part, either in the form of cells, tissue, or organs in aseptic culture conditions in viro. The most common problem causing failure in tissue culture activity is the happening of contamination on explants. Explants contamination can be minimized by using appropriate and effective sterilization methods. The aim of this research is to get the right and effective sterilization method in the control of contamination on the culture of callus of jeruk kacang (Citrus Reticulata L). This study use fours different sterilization methods, namely A, B, C and D method. Each treatment use the same disinfectant material, but different in conceration and duration of immersion. Each treatment group consists of 6 samples. The observation parameters in this study are the time of contamination and the number of contamination explants. The results of show that the method of sterilization D, immersion explants with alcohol 70% for 1 minute, Clorox 40% for 15 minutes, Clorox 20% for 15 minutes, HgCl2 0.05% for 3 minutes, which give the best result in dressing the amount of contamination of orange plants.
